Design of a Wounding Model for Bioengineered Skin Substitutes

open in viewerSkin provides the body's barrier function and therefore must heal quickly when wounded. Non-healing wounds require grafting or application of bioengineered skin substitutes(BES), which lack the ability to rapidly regenerate. In order to enhance the rate of regeneration of BES, there is a need for the development of an in vitro wounding model for BES, which is the goal of this project. A device was manufactured that created full-thickness, reproducible wounds to foreskin tissue, a model tissue for BES. Histology showed that a 90 second cryoburn from a liquid-nitrogen-cooled stainless-steel rod followed by a 72-hour culture period was successful in producing the desired wound. The result of this project is a model system and device that will be useful in the study of in vitro BES healing.
