Microscopy of Yeast and Bacteria

open in viewerInteractions between bacteria and fungi are a driver of many ecosystem functions. Bacteria have been shown to benefit from “fungal highways” as routes through the soil, though many aspects of these interactions are not well understood. Previous work has identified an interaction between the bacterial strain Pseudomonas putida and fungi Lyophyllum atratum. Here, we developed two methods to image both species individually and interacting. On glass slides, fungal hyphae were found to grow and were visualized with non-motile bacteria. On the oat flake agar surface, fungal hyphae did not grow individually, however, this system is promising for maintaining bacterial viability during imaging. This project builds the foundation for future studies to directly visualize bacteria-fungi interactions.
