Production of a putative mouse homolog for Protein AS160 and its use for antibody formulation for studying GLUT4 trafficking in adipocytes

GLUT4 trafficking is an integral process in insulin signaling. There are many different proteins involved, but a particular interesting protein is a substrate of Akt called AS160. AS160 was found to be insulin sensitive, being down-regulated in the cytosol, and up-regulated in low density microsomes. Working with Michael Czech at UMASS Medical Center, the focus of this MQP was to develop an antibody to mouse AS160 with a GST tag at the N-terminus. The antibody was then used with 3T3-L1 adipocytes as an in-vitro cell model to study the role of AS160 in GLUT4 trafficking.
