Taxus Cell Culture to Delivery System: A Novel Approach to Administer Paclitaxel

open in viewerPaclitaxel (PTX) is one of the leading chemotherapy drugs available for various solid tumor cancers and is most commonly administered systemically in a solution of dehydrated alcohol and Cremophor EL. This project investigated the potential for a Taxus suspension culture derived local delivery system of PTX. For this purpose, increased cell-associated PTX accumulation, DNA removal, and release kinetics were investigated. PTX was retained throughout a piloted decellularization process at an average of 0.68 mg/L with greater than 80% DNA removal. The decellularized biomass allowed for almost complete drug release within 72 hours of suspension. Initial release rate could be sustained when encapsulated in calcium alginate microbeads, suggesting this to be a feasible approach for PTX delivery.
