DNA MUTATION ANALYSIS OF THE VH4 INTRON LOCUS IN SMURF2 KNOCKOUT MICE

open in viewerThe Smurf2 gene in mice encodes a protein that mediates ubiquination and degradation of a germinal center transcription factor responsible for B-cell proliferation. Knockouts of the Smurf2 gene have been shown in our lab to increase the formation of Diffuse Large B-Cell Lymphomas. The purpose of this project was to clone and sequence the VH4 intron locus in the DNA from the germinal centers of spleens from Smurf2-KO mice to determine whether they have an increased DNA mutation rate prior to tumor formation. The cloning of the DNA was unsuccessful, indicating a need to conduct further experiments.
