Insertion of non natural amino acids
PublicThe scope of this report is to document the methodology used in creating nonsense codons in plasmid DNA by site directed mutagenesis as well as the application in site-specific incorporation of non-natural amino acids into target peptides. A technique to use artificial suppressor tRNA in an in vitro expression system based on a Micrococcus luteus S30 extract is suggested as an alternative to stop codon suppression. Site-specific protein mutagenesis has wide reaching implementations in biochemistry.
