Analysis of Subcellular Localization of PCV1-VP3 Using Mutant Variants

Porcine Circovirus 1 Viral Protein 3 (PCV1-VP3) selectively induces apoptosis in cancer cells. Localization within the cell is suspected to be a key factor in PCV1-VP3 inducing apoptosis. The purpose of this research is to analyze the subcellular localization of PCV1-VP3. Multiple mutants of the protein of interest were transfected into human non-small lung carcinoma cells (H1299), and transfected cells were analyzed using fluorescence microscopy and differential centrifugation. Results indicate that PCV1-VP3 associates with a perinuclear or nuclear structure. Comparison of wild type data to mutants supports the hypothesis that the nuclear export sequence (located a.a. 134-149) is significant in PCV1-VP3 localization.
