Incorporation of Tyrosine Kinase Receptor into a Hybrid Membrane for High Throughput Screening

open in viewerThis project focused on the development of a hybrid membrane for high throughput screening(HTS) for testing type II diabetes drugs. Self assembled monolayer(SAM) of dodecane thiol, octadecane thiol,and hexadecane thiol were created on gold and characterized by contact angle, ellipsometry, and impedance spectroscopy. Liposomes were created from the injection of dimyristoylphosphatidylcholine(DMPC) and TEM images were taken. Tyrosine kinase receptor, with no extracellular portion, was incorporated into these liposomes. A hybrid membrane of DDT and liposomes was formed and characterized by impedance spectroscopy and quartz crystal microbalance. Protein activity was detected with a kinase-glo assay. This hybrid membrane system can be used for HTS to test type II diabetes drugs.
