DIG-1 antibody purification
PublicThe protein DIG-1 is involved in the guidance of neuronal processes during the development of the nervous system in C. elegans. This MQP studied the purification of a previously raised antibody to a region in the C-terminus of the protein termed Con3. The antibody would be useful to locate the distribution of the protein in the worm to better understand its involvement in neuronal guidance. Experimental procedures included protein purification of Con3 from inclusion bodies of a baterial cell culture, IgG purification, dilution, and adsorption purification of serum from a rabbit injected with the protein, and embryo staining. The distribution of the protein during embryogenesis could not be determined by embryo staining, though future recommendations for this work include affinity purification of the antibody.
