Design and analysis of a cell lysate gravity settling system

A gravity-settling basis was built and analyzed to improve the efficacy of a continuous-flow aqueous two-phase electropartitioning apparatus. The initial design for the basin was based on theoretical fluid/particle flow dynamics. The performance of the basin was evaluated for both gel beads in water and E. coli cell lysate in a 4% dextran and 6% polyethylene glycol aqueous system. The results of our experimentation validate the use of such a settling basin for improving separation efficiency.
