Isolation and quantification of antigen specific T cells using magnetic activated cell sorting
公开Magnetic-activated cell sorting (MACS) was used to isolate and quantify antigen specific T cells. T cells specific to one vaccinia and three yellow fever peptides were isolated from two immune donors. Peptide specific cells were quantified by fluorescence-activated cell sorting analysis. Cytotoxic T lymphocyte (CTL) clones were generated from cultures obtained by the MACS assay through limiting dilutions. CTL assays on the bulk cultures and clones showed strong specific cytotoxic activity against target cells presenting their respective virus or peptide.
