Biochemical characterization of a protein-protein interaction

The plant, Arabidopsis thaliana, encodes a resistance gene, HRT, which is responsible for resistance to turnip crinkle virus (TCV). The goal of this project was to determine the amino acid properties required for recognition of the virus by carrying out specific site-directed mutagenesis on the TCV coat protein gene. Plants inoculated with the mutants demonstrated a tolerance for the virus, a situation that had never previously been observed in this model system.
