The Presence of DNA-binding protein LSF in a complex that binds to the switch region of immunoglobulin heavy chain gene gamma 3

This project was to search for a DNA binding protein, LSF, in the switch region upstream of the gene encoding heavy chain 3 (S3). LSF was previously shown to bind the switch regions of the heavy chain and [mu] genes. LSF has been shown to bind DNA and may regulate class switching. Class switch recombination is an altering of the somatic cell DNA to produce antibodies that recognize the same antigen, but have different effects within the immune system. Using a radiolabeled oligonucleotide of S3 in vitro binding of a protein was observed by EMSA. Further competition experiments supported the hypothesis that LSF bind to S3.
