The role of XRCC3 in homologous recombination

Interactions between DNA repair proteins Rad51 and XRCC3 were explored to investigate their roles in homologous recombination with respect to breast cancer. Recombinant plasmids and mammalian cell lines containing various XRCC3 mutants were constructed for experimentation with yeast hybrid and MMC sensitivity assays to test XRCC3 interaction with Rad51 or Rad51C in repair. Our results confirmed physical interactions between XRCC3 and Rad 51 or Rad51C, respectively. Mutations within or near the ATP-binding domain of XRCC3 almost always completely disrupted interaction with Rad51.
