Modification of a Collagen Scaffold to Enhance the Performance of Tissue Engineered Skin

There is a need for skin substitutes that provide enhanced vascularization and tissue ingrowth to the wound site to maximize skin regeneration and eliminate the need for additional surgeries. The team modified a novel full-thickness skin substitute known as micro-patterned dermal-epidermal regeneration matrix (μDERM) to enhance its ability to promote vascularization in vivo. Heparin and Fibroblast Growth Factor-2 (FGF-2) were incorporated into the dermal component. Migration of fibroblasts in response to these factors was assessed in vitro. While the non-heparinized sponges with a 300 ng/mL concentration of FGF-2 showed greatest outgrowth rates, the study would need to be performed over a longer period of time to understand the maximum potential effects of the heparin on the system.
