Analysis of Cytokine mRNA expression during the expansion of stem cell populations from human cord blood

Amplification of hematopoietic stem cells from human cord blood has applications for a variety of cell therapy protocols. The purpose of this MQP was to analyze the expression of various cytokines during the process of stem cell amplification in vitro. Total cellular RNA was isolated from pooled donated cord samples at day-0 or day-14 of amplification. cDNA probes were synthesized from the RNA samples, and hybridized to filter arrays containing equal quantities of 268 known cytokine DNA's to identify candidates for subsequent assay. The hybridization data indicated that neither day-0 or day-14 samples contained large quantities of inflammatory cytokine mRNA's, which facilitate subsequent perfusion into a patient. The expression of several developmentally interesting cytokine mRNAs was found to increase during stem cell amplification. Some of these upregulated mRNAs are known to be involved in stem cell growth.
