Mapping a nervous system gene in C. elegans -- a genetic linkage analysis of nu385

The nu385 mutation affects nervous system development in Caenorhabditis elegans. The mutant phenotype resembles the dig-1 mutation, which is suspected to be involved in cell adhesion during axon migration in neuronal development. The C. elegans strain containing the nu385 mutation, KP980, was crossed with a Hawaiian isolate containing numerous single nucleotide polymorphisms (SNPs). Many of these SNPs produce restriction fragment length polymorphisms, which provided physical markers for mapping the mutation by genetic linkage analysis. This method allows for easy identification of recombination events within single animals by PCR expediting the mapping process. Our results suggest nu385 is linked to markers on the right arm of chromosome IV, approximately 13 map units from the center. Ultimately, these results will lead to cloning and characterization of nu385.
